Cyclosporine is a cyclic polypeptide with 11 amino acids obtained from a fungus. This medicine specifically inhibits proliferation of T- lymphocyte cells, production of interleukin and cytokines. In cell mediated immunity antigen is processed and presented to helper T-cells. This secretes interleukins and cytokines that cause proliferation cytotoxic lymphocytes. The mature cytotoxic lymphocytes (killer cells) lyse antigen-containing cells. This medicine selectively suppresses cell-mediated immunity and prevents graft rejection. It suppresses the responses of primed helper T-cells also and hence used in the treatment of autoimmune diseases. It does not affect the bone marrow and reticuloendothelial system. It is more active when administered before antigen exposure.

Side effects

How does Cyclosporine works?

Cyclosporine inhibits cell mediated immunity system. The stimulation of T-cells produces a cascade of calcium dependent events and protein kinase activation. The calcium ions binding to calmodulin activate calcineurin, which dephosphorylates regulatory proteins. This medicine enters target cells and binds to a protein called cyclophilin. This complex then binds to and inactivates calcineurin thus inhibiting antigen stimulated activation and proliferation of T-cells.
